Lead halide perovskite nanocrystals (NCs) are important for future optoelectronic devices since they have a high photoluminescence quantum yield, show defect tolerance, and color tunability by their composition. The optical characteristics can be used in light emitting diodes (LEDs). However, the NCs need to fit simultaneously electronic requirements for their application in LEDs: long term stability under electrical bias, charge carrier balance, and facilitated charge injection are a prerequisite for their implementation. All three factors can be influenced by the organic ligand shell surrounding the NCs. By e.g. tight binding of ligands, the stability of the NCs can be improved. Hole and electron transfer rates can be tuned by ligand exchange and insulating surface ligands can be removed from the surface for easy charge transfer. Post-synthetic NC modification can lead to enhanced LED performance with reduced onset potential of luminance and high external quantum e!ciencies for extended time periods. Still, for building e!cient devices the underlying processes need to be understood and the energy barrier between the device layers determined. Therefore, charge injection of only one carrier needs to be studied depending on the surface chemistry of the NCs. Spectroelectrochemistry (SEC) is a method that helps understanding the role of surface ligands on charge injection into NCs. The combination of electrochemistry with photoluminescence (PL) spectroscopy makes the determination of the absolute valence (VB) and conduction band (CB) position possible. In addition, the effect of charging on the optoelectronic properties can be followed up.&#13;
In this cumulative dissertation, hole injection into CsPbBr3 NCs is studied&#13;
by PL SEC. The influences of the chemical environment on hole injection are&#13;
investigated. Thereby, the focus of this work lies on the modification of the ligand&#13;
shell and the self-assembly into supercrystals. The fundamentals of the examined&#13;
NCs and the method used for studying charge injection are introduced in chapter 2.&#13;
Chapter 3 uses PL SEC for investigating improved LED performance of didodecyldimethylammonium bromide (DDABr) covered NCs compared to as-synthesized NCs covered with oleic acid (OA)/oleylamine (OAm). Single carrier devices show an increase in hole current for DDABr NCs. In the PL SEC study, the VB of both ligands with different ligand densities is determined. Beforehand, ligand stripping is performed to create free surface sites for facilitated charge injection. Here, PL SEC shows that the VB position shifts to a lower potential for DDABr than OA/OAm NCs, while ligand stripping leads to an increased onset potential of VB doping for both ligand systems. The chapter highlights the improved charge carrier balance, resulting from promoted hole injection in LEDs for DDABr covered NCs caused by a VB shift of approx. 0.3 eV compared to OA/OAm covered NCs.&#13;
In chapter 4, inductive effects mediated by cinnamic acid (CA) derivatives on the electrochemical p-doping of CsPbBr3 NCs are studied. Protocols for the ligand exchange in solution are established in order to determine the hole injection potential for each system by PL SEC. The PL SEC study is supported by the performance of the CA-derivative covered NCs in LEDs and hole-only devices, which are in accordance with the reduced electrochemical hole injection potential for electron withdrawing compared to electron donating ligands determined by PL SEC. As this observation is in contrast to literature on PbS nanoparticles, the result is attributed to a kinetic effect. The comparison of the inductive effects introduced by the ligands highlight that the charge injection potential in LEDs is not only determined by the electronic structure, but can also be modified by a kinetic barrier introduced by the surface ligands.&#13;
In the final chapter of this thesis, electrochemical hole injection into supercrystals (SCs) is investigated. Here, the NCs are pretreated by ligand stripping and self-assembled into SCs. The spectroelectrochemical cell is placed on top of a confocal high-resolution microscope, which allows for spatial mapping of the SCs. The SCs show a high stability against the electrolyte solution and hole injection. The performance under electrical bias emphasizes the future application of SCs in LEDs.&#13;
In summary, the thesis shows that the hole injection potential can be tuned by 0.3 eV depending on the chemical environment of the CsPbBr3 NCs. Either by introducing DDABr as surface ligand or with electron withdrawing ligands, the hole injection potential can be tuned for a better alignment of the VB of the NCs with the hole transport layer in the LED. In addition, the assembly of NCs shows an increased stability after charge injection. The insights gained in this thesis demonstrate how charge carrier balance, and the long term stability under electrical bias as the electronic requirements of CsPbBr3 NCs can be tackled in future LED technology.

In dieser Arbeit wurde prospektiv die prognostische Rolle des EMID-Zeichens und verschiedener Biomarker auf die Erholung der LVEF bei PatientInnen mit vermuteter TCM untersucht.&#13;
Bei den eingeschlossenen Patienten handelte es sich um schwer kranke PatientInnen, mit im Mittel hochgradig reduzierter LVEF bei denen einen hochgradiger Verdacht auf das Vorliegen einer TCM bestand. Die Einschlusskriterien wurden dabei bewusst sehr eng gewählt, um einen möglichst guten Einblick in die zugrundeliegende Pathophysiologie zu bekommen. &#13;
Klinische Untersuchungen zeigten zunächst eine Verbesserung der mittleren linksventrikulären Ejektionsfraktion von rund 32% auf 45% sowie die Normalisierung der mittleren Herzfrequenz von 123/min auf 72/min nach Initiierung einer Rhythmuskontrolle.&#13;
Die histologischen Ergebnisse zeigten, dass rund die Hälfte der PatientInnen ein mittelstarkes bis stark ausgeprägtes EMID-Zeichen aufwies.&#13;
Darüber hinaus konnte die Hypothese bestätigt werden, dass die Ausprägung des semiquantitativ evaluierten EMID-Zeichens mit dem Schweregrad der Erkrankung negativ korreliert: Je stärker das EMID-Zeichen ausgeprägt war, desto schlechter war die Entwicklung der PatientInnen hinsichtlich der Erholung der LVEF.&#13;
Des Weiteren konnte die Hypothese, dass die bei Einschluss gemessenen Biomarker mit dem Schweregrad der Erkrankung, gemessen am ∆ der LVEF von Einschluss zu follow-up, korrelieren, teils bestätigt werden. Insbesondere das C-reaktive Protein konnte hier als prognostischer Marker identifiziert werden. &#13;
Nach Aufteilung in zwei Gruppen je nach Erholung der LVEF (∆LVEF ≤ 15% und ∆LVEF &gt; 15%) konnten für die sich schlechter erholende Gruppe signifikante Veränderungen in den Plasma-Konzentrationen für Osteopontin (OPN) und Myeloperoxidase (MPO) beobachtet werden. Die hier gemessenen Konzentrationsunterschiede entwickelten sich jedoch zum Teil anders als dies für andere Kardiomyopathien beobachtet wurde. Somit wird die Vermutung bekräftigt, dass es sich bei der TCM um eine eigenständige Kardiomyopathie mit eigener Pathophysiologie und Risikofaktoren handelt, welche sich in Diagnostik und Therapie maßgeblich von anderen, „klassischen“ Kardiomyopathien unterscheidet. &#13;
Zukünftige Studien mit höheren Fallzahlen sind erstrebenswert, um die Aussagekraft der einzelnen Ergebnisse zu stärken und eine detailliertere Risikostratifikation für den klinischen Alltag zu erarbeiten.

Die Entdeckung der Antibiotika war ein Meilenstein in der Geschichte der Medizin. Sie führten zu einer erheblichen Verringerung der Sterblichkeitsraten und ermöglichten durch die Kontrolle infektiöser Komplikationen die Entwicklung invasiver medizinischer Behandlungen. Ihre weit verbreitete Verwendung hat jedoch auch die weltweite Entwicklung von Resistenzen gefördert, die heute eine ernsthafte Bedrohung für die globale Gesundheit darstellen. Besonders besorgniserregend sind multiresistente Gram-negative ESKAPE-Erreger wie Pseudomonas aeruginosa. Dieser verursacht schwere nosokomiale Infektionen mit hohen Sterblichkeitsraten bei immungeschwächten Patienten. &#13;
Zu den wichtigsten Mechanismen der β-Lactam-Resistenz von P. aeruginosa gehören die Inaktivierung von Antibiotika durch die Überexpression der chromosomalen β-Lactamase AmpC, eine verringerte Permeabilität der äußeren Membran, ein erhöhter Efflux durch RND-Transporter sowie Veränderungen der Zielproteine, also der Penicillin-bindenden Proteinen. Unter dem Druck von Antibiotika können auch nicht-essenzielle Gene essenziell werden. Die gezielte Hemmung der entsprechenden Proteine durch Adjuvantien könnte die Empfindlichkeit resistenter Bakterien gegenüber Antibiotika wiederherstellen. In P. aeruginosa wurden die LytM/M23-Zink-DD-Endopeptidasen MepM1, MepM2 und MepM3 als vielversprechende Wirkziele einer Adjuvantien-Therapie identifiziert, da ihre Deletion die Resistenz gegenüber verschiedener bekannter und gut verträglicher β-Lactam-Antibiotika synergistisch bricht. Darüber hinaus teilen sich MepM1 und MepM2 das gleiche aktive Zentrum, sodass beide Enzyme mit einem einzigen Hemmstoff inhibiert werden könnten. &#13;
Diese Arbeit konnte zeigen, dass eine verringerte β-Lactamase-Aktivität sowie überraschenderweise erhöhte MexE/MexF/OprN-Spiegel verantwortlich sind für die Verringerung der Resistenz gegen β-Lactam-Antibiotika nach Deletion von mepM. Es wird angenommen, dass erhöhte MexE/MexF/OprN-Spiegel das Quorum Sensing, die Motilität oder die Biofilmbildung beeinflussen und damit die Antibiotikaresistenz reduzieren können. Massenspektrometrische Analysen ergaben weitere potenzielle Veränderungen des Membranproteoms durch die Deletion von mepM1, mepM2 und mepM3. Dies brachte neue potenzielle Resistenzmechanismen und Funktionen von MepMs auf. Hierzu zählt die Anpassung an veränderte Umweltbedingungen durch Nährstoffaufnahme, Quorum Sensing, Motilität, Biofilmbildung und Umbau der Zellhülle. Die erfolgreiche Entwicklung von Aufreinigungsprotokollen für die rekombinanten, aktiven Enzyme PaMepM1 und PaMepM2 sowie die Herstellung von Anti-MepM-Antikörpern stellen wichtige Ressourcen für die zukünftige Forschung dar. Die Antikörper wurden zur Validierung der ∆mepM1- und ∆mepM2-Deletionsmutanten verwendet und bestätigten, dass MepM1, im Gegensatz zu MepM2, durch CtpA reguliert wird. Durch die Entwicklung funktioneller Enzymaktivitätstests konnte erstmals die Aktivität der Murein-DD-Endopeptidasen von P. aeruginosa in vitro gemessen werden, was den Weg für eine weitere enzymatische Charakterisierung ebnete. Schließlich wurde ein Hochdurchsatz-Assay zum Testen potenzieller MepM-Inhibitoren entwickelt. Die Identifizierung der ersten hemmenden Verbindung beweist die Funktionalität des Versuchsaufbaus. Angesichts der dringenden Notwendigkeit, multiresistente P. aeruginosa-Infektionen zu bekämpfen, leistet diese Arbeit einen wertvollen Beitrag zur Erforschung der Antibiotikaresistenz und zur Entwicklung neuer antimikrobieller und antivirulenter Strategien.; The discovery of antibiotics was a milestone in the history of medicine. They led to a significant reduction in mortality rates and enabled invasive medical treatments to be developed by controlling infectious complications. However, their widespread use since then has encouraged the worldwide development of resistance, which today poses a serious threat to global health. Of particular concern are multidrug-resistant Gram-negative ESKAPE pathogens, such as Pseudomonas aeruginosa, which is associated with severe nosocomial infections and high mortality rates in immunocompromised patients. Key mechanisms of P. aeruginosa β-lactam resistance include antibiotic inactivation via the overexpression of the chromosomal β-lactamase ampC, reduced outer membrane permeability, increased efflux through RND transporters and target modifications in penicillin-binding proteins. Under antibiotic pressure, non-essential genes can become essential. Targeting the corresponding proteins with adjuvants could restore antibiotic susceptibility in resistant bacteria. The LytM/M23 zinc DD-endopeptidases MepM1, MepM2 and MepM3 have been identified as promising adjuvant targets in P. aeruginosa, as deleting them breaks resistance synergistically against various well-known and well-tolerated β-lactam antibiotics. Additionally, MepM1 and MepM2 share the same active site, enabling both enzymes to be targeted simultaneously with a single inhibitor. &#13;
This work demonstrated that reduced β-lactamase activity and, surprisingly, increased MexE/MexF/OprN levels are responsible for the reduced resistance to β-lactam antibiotics observed following the deletion of mepM. It is hypothesised that elevated MexE/MexF/OprN levels may affect quorum sensing, motility, or biofilm formation, thereby influencing antibiotic resistance. Analyses using mass spectrometry revealed further potential changes of the membrane proteome following the deletion of mepM1, mepM2 and mepM3. This brought up new potential resistance mechanisms and functions of MepMs including adaptation to altered environmental conditions through nutrient uptake, quorum sensing, motility, biofilm formation and remodeling of the cell envelope. The successful development of purification methods for the recombinant, active enzymes PaMepM1 and PaMepM2, as well as the generation of anti-MepM antibodies, provides important resources for future research. These antibodies were used to validate the ∆mepM1 and ∆mepM2 deletions, confirming that MepM1 is regulated by CtpA, in contrast to MepM2. The development of functional enzyme activity assays made it possible to measure P. aeruginosa murein DD-endopeptidase activity in vitro for the first time, paving the way for further enzymatic characterization. Finally, a high-throughput assay for testing potential MepM inhibitors was developed, representing a promising step towards the discovery of therapeutic agents that target MepMs. The identification of the first inhibitory compound proves the functionality of the experimental design. Considering the urgent need to combat multidrug-resistant P. aeruginosa infections, this work makes a valuable contribution to research into antibiotic resistance and the development of new antimicrobial and antivirulence strategies.

Neuere Fortschritte in der Nickelkatalyse haben den synthetischen Werkzeugkasten für die Bildung von C–C- und C–Heteroatom-Bindungen erheblich erweitert. In dieser Arbeit werden fünf Studien zusammengeführt, die zeigen, wie sich Katalysatordesign, mechanochemische Aktivierung und mechanistisches Verständnis zusammenfügen lassen, um lang bestehende Einschränkungen in der Übergangsmetall-katalysierten Synthese zu überwinden. Ein zentrales Thema dieser Dissertation ist die Entwicklung effizienter nickelkatalysierter Transformationen unter milden und operativ einfachen Bedingungen bei gleichzeitiger Aufklärung der katalytisch aktiven Spezies, sowie deren Deaktivierung und Regenerierung.
